Sou farmacêutico e possui mestrado em Farmacologia na Faculdade de Farmácia, Universidade da Uppsala (1968) e doutorado em Farmacologia ? da mesma universidade (1978). Trabalhei como pesquisador na companhia Pharmacia em Uppsala durante 12 anos do que 6 anos em Departamento Medicina Experimental especializada em estudos da microcirculação e em seguida como pesquisador em farmacologia e microcirculação na companhia AstraZeneca em Lund durante 12 anos. Atualmente estou professor visitante do Instituto Biofísica Carlos Chagas Filho da Universidade Federal do Rio de Janeiro atuando principalmente nos seguintes temas: microcirculação e estudos com microscopia intravital sobre efeitos agudos das parasitas (Trypanosoma cruzi, Leishmania chagasi) e das homogenatos das glândulas salivar da Lutzomyia longipalpis e o peptídeo vasodiladora, maxadilan, que foi isolado da glândula salivar. Também estou estudando os efeitos das bactérias (Porphyromonas gingivalis) na microcirculação e a ativação do sistema imune inata e o papel de cininas (bradicinina). (Texto informado pelo autor)

Edema Inflamatório na bolsa bochecha do hamster: um modelo de estudo sobre a inter-relação funcional entre mastócitos e o sistema de contato (calicreína-cinina).
Descrição: Integrada por 3 serino proteases plasmáticas (FXII, PK e FXI) e uma proteína adaptadora não enzimática (cininogênio de alto peso molecular [HK]), a via de contato de coagulação, freqüentemente referida como sistema calicreína-cinina (SCC), foi recentemente implicada na patogênese de diversas doenças trombo-inflamatórias experimentais, inclusive no acidente vascular cerebral (AVC). Considerando que as seqüelas do AVC foram mitigadas em camundongos transgênicos deficientes tanto em Fator XII quanto HK, os iniciadores do SCC, estes estudos sugerem que inibidores seletivos de FXII poderão ter efeitos terapêuticos, atenuando/prevenindo (i) trombose (ii), o edema inflamatório provocado pela bradicinina, um nonapeptídeo pró-inflamatório liberado de HK pela ação da PKa. Trabalhos recentes em um novo modelo de infecção chagásica (parasitas inoculados pela via intramiocárdica) sugerem que os tripomastigotas (T.cruzi Dm28c) induzem edema intersticial intramiocárdico e invadem células cardíacas através de mecanismos que dependem da ativação interdependente de receptores de bradicinina (BKRs) e endotelinas (ETRs) (Andrade et al., SBPz, 2012). Estudos de microscopia intravital (modelo da bolsa da bochecha, HCP) indicam que tripomastigotas geram bradicinina através de mecanismos proteolíticos potencializados por histamina secretada por mastócitos perivasculares (Nascimento et al., SBPZ, 2012). No presente projeto, pretende-se verificar se inibidores do FXIIa e PKa (1) reduzem o edema inflamatório induzido por tripomastigotas em HCP (normal versus inflamado) (2) verificar se estabilizadores de mastócitos (cromoglicato de sódio) são capazes de reduzir/prevenir o edema inflamatório e a deposição de fibrinas no sítio de infecção chagásica (3) comparar o fenótipo pró-inflamatório de Dm28c DTU I) com os fenótipos das cepas VL-10 e 115 (respectivamente DTU V e VI). (4) verificar se inibidores de FXII e PKa inibem extravasamento de plasma induzido pelo periodontopatógeno Porphyromonas gingivalis. CRONOGRAMA POR OBJETIVOS ESPECÍFICOS I.Usar inibidores de bradicinina (HOE-140), e de endotelina (Bosentan) e de degranulação das mastócitos (Cromoglicato de Sódio) em experimentos com T. cruzi Dm28c aplicado na bolsa normal (HCP) ou inflamado (com uma pequena dose de histamina). II.Usar cepas diferentes (T. cruzi Dm28c, VL-10, e 115, respectivamente DTU I, V e VI) para investigar se a ativação extravascular de SCC pode ser dependente da cepa. III.Usar um substrato sintético (S-1125), que na ativação do SCC pode formar peptídeos fluorescentes (mostrando formação da cininas ou BK), que pode ser medido na solução de superfusão da bolsa (HCP) no mesmo momento que o extravasamento é medido, com análise de imagens por microscopia intravital. IV.Usar um inibidor seletivo de Fator XII (peptídeo) com eficácia comprovada in vitro (usando substrato sintético S-1125) e um inibidor da calicreína (PKa) e aplicar isso antes da aplicação de histamina e TCTs na bolsa da bochecha (HCP). Mecanismos envolvidos no aumento da permeabilidade vascular por lisados de glandula salivar de Lutzomyia longipalpis e promastigotas de Leishmania chagasi
Descrição: Estudos recentemente realizados pela nossa equipe demonstram que promastigotas metacíclicos de espécies pertencentes ao complexo Leishmania donovani (L. donovani e L. chagasi) provocam aumentos rápidos da permeabilidade microvascular (APMV) em hamsters e camundongos, devido a sua capacidade de ativar o sistema cinina mediante ação de uma cisteína-protease parasitária (Svensjö et al. 2006) [9]. Além de envolver a participação do receptor B2 de bradicinina, experiências recentes realizadas no modelo da bolsa da bochecha de hamsters sugerem que o APMV também depende da ativação do receptor H1 de histamina. Observações adicionais sugerem que a aplicação tópica dos parasitas na bolsa da bochecha promove rapidamente a aderência de neutrófilos ao endotélio. Não obstante os progressos realizados no estudo dos mecanismos envolvidos no APMV induzido pela L. chagasi, é razoável prever que na infecção natural do homem, os efeitos vasculares e imunológicos induzidos pelos parasitas devem ser sobrepostos aos efeitos biológicos da saliva do inseto transmissor. Em busca de modelos experimentais que possam simular este complexo cenário, recentemente iniciamos estudos para caracterizar a APMV induzida por lisados de glândulas salivares do inseto vetor Lutzomyia longipalpis (SGL). Nossos estudos indicam que SGL também induz APMV na bolsa da bochecha do hamster. Os estudos que fazem parte deste plano objetivam caracterizar as vias de ativação celular implicadas na APMV induzida por aplicações (separadas e conjuntas) de SGL e promastigotas. Na primeira etapa deste projeto, pretendemos caracterizar a seqüência temporal de eventos subjacentes ao APMV induzidos por promastigotas e por SGL. Pretendemos determinar se (i) existe uma relação causal entre ativação de neutrófilos e APMV induzida pelos parasitas e (ii) caso isto seja comprovado, investigaremos o papel de cininas e da anafilatoxina C5a na APMV. Na segunda etapa, estudaremos os mecanismos envolvidos na indução de APMV por lisados de glândula salivar de L. longipalpis, com ênfase na participação de maxadilan, uma substância vasodilatora bem caracterizada, sabidamente capaz de estimular inflamação mediante ativação de receptores PAC1 presentes em neutrófilos. III. OBJETIVOS ESPECÍFICOS1.Avaliar o papel de neutrófilos no APMV (aumentos rápidos da permeabilidade microvascular) da bolsa da bochecha do hamster topicamente exposta a promastigotas metacíclicos L. chagasi.2.Tal como descrito acima, na presença de SGL ou maxadilan.3.Verificar se a ativação do sistema complemento é importante para indução da (i) marginação de leucócitos (ii) indução de APVM no leito vascular exposto aos promastigotas4.Caso haja indício da participação do sistema Complemento, verificar se o APMV dependente de ativação de receptores H1 é mediado pela anafilatoxina C5a. 5.Caracterizar os mecanismos envolvidos no APMV induzidos por lisados de glândula salivar de Lutzomyia longipalpis, bem como de maxadilan recombinante e também em combinação com promastigotas.6.Investigar a participação de óxido nítrico no estado de refratariedade vascular induzida pela primeira aplicação de parasitas no leito vascular.Metas Específicas:1.Estabelecer métodos quantitativos de detecção de marginação e aderência de neutrófilos em veias pós-capilares da bolsa da bochecha do hamster (BBH), após aplicação tópica de promastigotas, na presença ou ausência de SGH.2. Usando o fator anti-complementar do veneno de cobra, determinar se a marginação e a aderência de leucócitos, e APMV, dependem da ativação do sistema complemento pelos parasitas. 3. No mesmo modelo, verificar se a marginação e ativação de leucócitos, e APMV, é dependente de liberação de histamina. 4.No mesmo modelo, verificar se a depleção de Complemento in vivo através do tratamento dos hamsters com Veneno de Cobra [43] reduz a marginação de leucócitos e/ou APMV. 5. Caso a resposta.
