2012 - 2012 | Technical training on collecting bees and molecular analysis. |
Description: This project aims to provide to the student the opportunity of gathering experience on collecting bees and analysing data by helping postgraduate student to develop their research. The student will be responsible for collecting Tetragonisca angustula, Bombus morio and Bombus pauloensis for two PhD projects. He will also be involved in the data analysis and laboratory experiments related to both projects. Thereat, the student will obtain training on population genetics data analysis and laboratory techniques, such as cloning, bacterial transformation, sequencing, DNA and RNA extraction, etc..

2010 - 2013 | Genetic variability of Tetragonisca angustula (Hymenoptera, Apidae, Meliponini) from meliponaries. |
Description: Low genetic variability has normally been considered a consequence of animal husbandry as well as a major contributing factor to bee decline. Here we performed molecular analysis of captive populations of the stingless bee Tetragonisca angustula, one of the most commonly kept species across South America. We sampled 430 individuals, one per nest, from nine distinct meliponaries (same as apiaries, but for stingless bees) in Sao Paulo and Parana states, Brazil. As positive control for genetic variability, we reanalyzed data from wild samples reported in previous studies. Employing microsatellite markers, we verified a similar genetic variability between wild and captive populations. Nonetheless, genetic variability differences at mitochondrial DNA were remarkable between populations. Gene flow by males, transport of nests and division of colonies are the most probable explanations for the observed pattern of genetic structure. We conclude that increasing the number of colonies kept through nest divisions does not negatively affect nuclear genetic variability, which seems to be maintained by local male dispersal and human-mediated nest transport..

2008 - 2010 | Technical training on collecting bees and molecular analysis. |
Description: Natural stingless bees populations are poorly known, which means that the impact of habitat fragmentation on those population cannot be yet evaluated properly. Recent techniques on molecular genetics allows us to get a proper estimative of the genetic variability of existing population and how large they should be to be genetically healthy. The comprehension of those processes is crucial for every effort toward conservation. The laboratory of genetics and evolution of bees, which uses molecular markers to better understand the systematics and population dynamics of stingless bees, have initiated research on this direction. In University of Sao Paulo campus in Ribeirao Preto city, and surrounding natural environment, about 1025 were located and are constantly monitored. This project aims to perform the same type work in the University of Sao Paulo, Sao Paulo campus, which will represent a small population in anthropic area. United, both data will allow us to perform population comparisons, which will improve our understanding on the biology of stingless bees. We will get both information about gene flow, genetic variability in different environmental and captive conditions and possible evidence of speciation processes. The application of the acquired knowledge will increase our understanding about stingless bee beekeeping and how to preserve those species properly..

2004 - 2005 | Survey of the species diversity and characterisation of the mitochondrial DNA of bees from Parque Ecológico da Cachoeirinha, Goias state, Brazil. |
Description: Bees are dependent on flower nectar and pollen (main sources of protein and energy, respectively) and have an important economic and ecological role as pollinators, representing about 40 to 90% of phanerogams pollinators in several ecosystems. This work presents a description of bee species from Parque Municipal da Cachoeirinha in Iporá, Goiás state, Brazil. In order to have an overall idea of the genetic diversity of those bees, we also performed a molecular analysis through PCR+RFLP of the mitochondrial DNA in six species. A total of 58 hours of capturing were performed on November and December 2004 and on March and May 2005. Nine hundred thirty-three individuals were collected, distributed in one family, 16 tribes, 33 genera, and 62 species. Meliponini tribe presented the highest number of individuals and species. Most abundant species were Trigona chanchamayoensis (16.51%), Tetragona clavipes (14.79%) and Apis mellifera and Trigona recursa (12.33%). By using three restriction enzymes on mitochondrial DNA fragments amplified via PCR, 5 distinct haplotypes were found among the six species, indicating moderate genetic variability. This work presents the first occurrence in Goiás state for six genera and 12 species..
